I added a property that is checking if the lead having relation to a conference, the answer are fixed to yes or no, How can I get new property with the option to write only when the answer is yes?

As far as I understand, if the tag is yes, then the automation has to go to other database and select the correct related conference and then add it to that conference property.
It is not possible to add database relations like that using Notion automations.
If you already have a relation to a conference, why not use that property instead?
You do not even need a tag property to change it to Yes or No, just create a new relation property for the conference and relate them as needed, if there is something related it appears, if not, then not.
Otherwise you will have to go with formulas and create a formula to show relations if the selector is set to Yes, otherwise show something else or nothing.
